What’s intriguing is that the science of aging has made remarkable strides in recent years, with human trials on the horizon. People born in the U.S. today can expect to live to an average age of about 79. A century ago, life expectancy was closer to 54. “We’ve had a significant increase in lifespan over the last century,” says Dr. Marie Bernard, deputy director of NIH’s National Institute on Aging.
1. Senescent Cells: The “Zombie Cells”
Our cells bear an expiration date. With each cell division, chromosomes are copied, gradually losing fragments of DNA at the ends. To safeguard against potential catastrophe, cells feature lengthy DNA segments known as telomeres, akin to the stiff bits on shoelaces.

Older mice lacking senescent cells exhibited heightened activity levels, improved cardiac and renal function, and a reduced cancer risk. Not only did they outlive their average counterparts by up to 30%, but they also enjoyed superior health.
Given the impracticality of genetically modifying all human cells, the challenge lies in devising methods to eliminate senescent cells without harming their healthy counterparts. It turns out that senescent cells produce insufficient levels of a protein that signals their programmed death.
In a late 2016 study, mice were administered this protein via injection, resulting in the demise of 80% of their senescent cells with minimal repercussions for healthy cells. Treated mice experienced overall health improvements, even witnessing hair regrowth. Consequently, numerous companies are exploring therapies targeting senescent cells, with human trials imminent.

Regrettably, NAD+ cannot be ingested in pill form as it struggles to enter cells directly. However, scientists have identified more adaptable substances capable of penetrating cells and converting into NAD+. In 2016, multiple trials involving mice demonstrated that these substances rejuvenated skin, brain, and muscle stem cells, enhancing DNA repair capabilities and slightly extending lifespans. This revelation even piqued the interest of NASA, exploring strategies to mitigate DNA damage caused by cosmic radiation during missions to Mars.
While human trials are in the pipeline, it’s too early to definitively state whether these interventions will extend our healthspan or lifespan. Nevertheless, NAD+ emerges as a strong contender for becoming the inaugural human anti-aging pill.
3. Stem Cells: The Architects of Renewal
Stem cells, often likened to the architectural blueprints of our cells, are dispersed throughout the body. Another study transplanted embryonic mouse stem cells directly into the hearts of older mice, resulting in enhanced cardiac function, increased endurance, and even accelerated hair regrowth.
These discoveries underscore the intricacies of aging, highlighting that there is no singular “magic bullet” to combat the rigors of growing old. Instead, a blend of diverse therapies may hold the key. The combination of senescent cell clearance, stem cell replenishment, and metabolic regulation through medication presents a comprehensive approach to promoting healthier aging.
It is essential to acknowledge that while these studies have displayed promise in mice, the extent of their success in humans remains uncertain. Nevertheless, they serve as a proof of concept. To fully grasp how we can extend our healthspan, human trials are imperative. This article has merely scratched the surface of the myriad ongoing research endeavors in this field. The realm of healthspan extension deserves heightened attention and funding, as it holds the potential to alleviate the burdens of aging and enable us to grow old without enduring pain.
